摘要

目的:观察氟西汀联合毛蕊花糖苷(Act)对抑郁症大鼠CREB 表达、学习记忆能力的影响。方法:将SD 大鼠随机分为对照组( 健康大鼠)、模型组( 抑郁症模型大鼠)、氟西汀组( 模型+ 氟西汀)、Act 组( 模型+ 毛 蕊花糖苷)、联合组( 模型+ 氟西汀+ 毛蕊花糖苷),分别采用旷场实验、糖水测试观测抑郁程度,水迷宫、平衡 木实验观察学习记忆能力,免疫印迹检测大鼠海马组织中CREB 蛋白表达。结果:各组大鼠实验1 d 时无差异; 在经过氟西汀与毛蕊花糖苷的干预后,随着时间的增加,大鼠水平活动、站立次数、糖水偏好增加,逃避潜伏期 与通过平衡木时间降低,且以联合组效果最显著。免疫印迹结果显示,与对照组相比,模型组CREB 水平降低, 在经过氟西汀与毛蕊花糖苷的干预后CREB 水平增加,且以联合组效果更显著。结论:氟西汀联合毛蕊花糖苷可 改善大鼠抑郁行为,可提高CREB 表达和学习记忆能力。

Abstract

Objective : To observe the effects of fluoxetine combined with acteoside (Act) on CREB expression, and learning and memory ability in depressed rats. Methods : SD rats were randomly divided into a control group (healthy rats), a model group (model rats with depression), fluoxetine group( model rats with fluoxetine), an Act group (model rats with acteoside) and a comined group (model+fluoxetine+acteoside). Open field experiment and the sugar water test were used to judge respectively the degree of depression. Water maze and balance beam experiments were performed to observe learning and memory ability. Western blotting was performed to detect CREB expression in rat hippocampus. Results : After fluoxetine and acteoside intervention, the level of activity, standing times, sugar water preference of rats increased with the increasing of time, escape latency and passing balance beam time decreased, and the combined group showed the most significant changes. Western blotting results showed that CREB level was decreased in the model group, and increased after the fluoxetine and acteoside intervention, and the changes were more significant in the combined group. Conclusion : Fluoxetine combined with acteoside can improve depression behavior, CREB expression and learning and memory ability of rats.
